Transformation through somatic Embryo of Oenanthe stolonifera with agrobacterium


Abstract
A gene transformation system was established for water dropwort (Oenanthe stolonifera DC) by infecting somatic embryos with Agrobacterium tumefaciens containing a binary vector pBI121. Transformation efficiency was examined by comparing two systems for regeneration. Somatic embryos were cocultivated with Agrobacterium in liquid MS medium containing, in system [A], 1 §·/ §¤ 2,4-D, that resulted in production of embryogenic callus and subsequently embryos when tissues were cultured on a 2,4-D free medium, or in system [B], 2.5 §·/ §¤ BA, which resulted in the production of secondary embryos. System [B] produced a higher transformation frequency primarily as a result of greater embryo production. Transformation and stable insertion of vector DNA in the host genome was comfirmed by ¥â-glucuronidase (GUS) expression as determined by both fluorometric and histochemical assays and by DNA blot and polymerase chain reaction (PCR) analysis. Several transgenic plants were produced by repetitive secondary embryogenesis of transfomed mother embryos.
